Case history: pertussis toxin

• Pertussis toxin is heterohexamer of 105kDa

  • room for 2 or 3 in cell

• Poor isomorphism: phases good to about 6Å

• No peaks in self-rotation or native Patterson

• Heavy atoms do not obey NCS

• Found molecules in Wang envelope

• Determined NCS operators with real-space molecular replacement

• Determined envelope from local correlation map
